目的 :利用医学研究常用ICR品系小鼠,在建立窦状卵泡体外培养体系的基础上,研究黄体生成素(luteinizing hormone,LH)与表皮生长因子(epidermal growth factor,EGF)诱导卵母细胞体外成熟及排卵的作用。方法 :体外剥离得到大的窦状卵泡用于培养并建立适宜于研究诱导卵母细胞成熟及排卵的窦状卵泡体外培养模型,通过向培养液中添加LH和EGF诱导卵母细胞体外成熟与排卵。结果:成功构建了ICR小鼠窦状卵泡体外培养模型,发现LH或EGF单独处理均可诱导卵泡卵母细胞成熟和卵丘扩展,而两者共同作用才能够诱导卵泡体外排卵。结论:LH/EGF促进ICR小鼠窦状卵泡卵母细胞体外成熟与排卵。
OBJECTIVE: To study the effects of luteinizing hormone (LH) and epidermal growth factor (EGF) on the maturation of oocytes in vitro based on the establishment of an in vitro culture system of ICR strains. And the role of ovulation. Methods: A large sinusoid follicle was dissected in vitro for culturing and establishing an in vitro culture model of sinusoid follicles suitable for studying the induction of oocyte maturation and ovulation. The in vitro maturation and ovulation of oocytes were induced by adding LH and EGF to the culture medium . Results: The model of sinusoidal follicles in ICR mice was successfully established. It was found that follicular oocyte maturation and cumulus expansion could be induced by LH or EGF alone. However, both of them could induce follicle ovulation in vitro. Conclusion: LH / EGF promotes the in vitro maturation and ovulation of antral follicular oocyte in ICR mice.
