目的探讨人脑胶质瘤中11 C-蛋氨酸(11 C-MET)摄取量与细胞增殖和血管形成的关系。方法对30例新诊断的脑胶质瘤患者行11 C-MET PET检查,计算11 C-MET最大标准化摄取值(SUVmax);采用免疫组化方法检测Ki-67和CD34抗原在相同脑胶质瘤标本中的表达,计算Ki-67标记指数(Ki-67LI)和微血管密度(MVD)。结果 11 C-MET SUVmax和Ki-67LI均随胶质瘤病理级别的升高而明显增加(P=0.000,P=0.000);在高恶性度胶质瘤中11 C-MET SUVmax和Ki-67LI均明显高于低恶性度胶质瘤(P=0.000,P=0.000);而MVD在胶质瘤不同病理级别间(P=0.831)以及高、低恶性度胶质瘤间(P=0.370)均无明显差异。11 C-MET SUVmax与Ki-67LI存在明显正相关(P=0.000),而11 C-MET SUVmax与MVD(P=0.154)以及Ki-67LI与MVD(P=0.842)之间均无明显相关性。结论 11 C-MET摄取量和细胞增殖活性可较好地反映胶质瘤病理级别和恶性程度。
Objective To investigate the relationship between 11 C-methionine (MET) intake and cell proliferation and angiogenesis in human gliomas. Methods Eleven newly diagnosed patients with gliomas underwent 11 C-MET PET examinations to calculate the maximum normalized 11 C-MET uptake value (SUVmax). Immunohistochemistry was used to detect the expression of Ki-67 and CD34 on the same brain glia The Ki-67 labeling index (Ki-67 LI) and microvessel density (MVD) were calculated from the tumor specimens. Results Both 11 C-MET SUVmax and Ki-67LI increased with the pathological grade of glioma (P = 0.000, P = 0.000) (P = 0.0007, P = 0.000), but MVD was significantly higher in gliomas with different pathological grades (P = 0.831) and between high and low grade gliomas (P = No significant difference. 11 C-MET SUVmax had a significantly positive correlation with Ki-67LI (P = 0.000), while there was no significant correlation between 11 C-MET SUVmax and MVD (P = 0.154) and Ki-67LI and MVD . Conclusions 11 C-MET uptake and cell proliferation activity can well reflect the pathological grade and malignant degree of gliomas.
