设置7个萃取温度(25,35,45,55,65,75,85℃)和萃取时间(30,40,50,60,70,80,90min)来选择超声提取紫椴花中多糖的最佳提取温度和提取时间。利用分光光度法对紫椴花中的多糖含量进行了分离和测定。结果表明:最佳提取条件为提取温度为75℃,提取时间为52min。经蒽酮--硫酸显色,于580nm处测定,其多糖的含量为9.74%,RSD=0.47%(n=3)。此测定方法简便,样品溶液在4h内显色稳定,重现性较好,平均回收率为99.48%-1.01%,RSD=0.112%(n=3)。图1表3参13。
Seven extraction temperatures (25, 35, 45, 55, 65, 75 and 85 ℃) and extraction time (30, 40, 50, 60, 70, 80 and 90 min) Good extraction temperature and extraction time. The polysaccharide content of Tilia amurensis was separated and determined by spectrophotometry. The results showed that the optimum extraction conditions were extraction temperature of 75 ℃ and extraction time of 52 min. The anthrone-sulfuric acid color was measured at 580nm, the polysaccharide content of 9.74%, RSD = 0.47% (n = 3). The determination method is simple, the color of the sample solution is stable within 4 hours and the reproducibility is good. The average recovery rate is 99.48% -1.01%, RSD = 0.112% (n = 3).
