目的:研究银杏内酯类合成物F(简称药物F)同时拮抗组织胺(HA)和血小板激活因子(PAF)的作用。方法:测定离休豚鼠肺组织的收缩率以及用药物F后的致敏豚鼠肺功能的变化。结果:(1)加入药物F后离体豚鼠的气管条和肺条对HA的收缩率分别由(50.97±16.00)%和(54.2±12.17)%降至(21.69±3.85)%和(22.97±17.78)%(P<0.05);(2)药物F保护后,离体豚鼠肺条对PAF的相对收缩率由(89.49±16.00)%降至(46.21±14.23)%(P<0.05);(3)药物F保护的豚鼠致敏后,其肺功能较对照组有改善趋势。结论:药物F能同时拮抗离体豚鼠肺组织对HA和PAF的收缩反应,并有改善致敏豚鼠肺功能的趋势,可望用于支气管哮喘的治疗。
Objective: To study the effects of ginkgolide compound F (abbreviated as drug F) on the antagonistic activity of both histamine (HA) and platelet activating factor (PAF). METHODS: The contractile rate of lung tissue in isolated guinea pigs and changes in lung function of sensitized guinea pigs after administration of drug F were measured. RESULTS: (1) After the addition of drug F, the contraction rates of HA and tracheal strips of isolated guinea pigs decreased from (50.97±16.00)% and (54.2±12.17)%, respectively. (69±3.85)% and (22.97±17.78)% (P<0.05); (2) The relative contraction rate of PAF in isolated guinea pig lung strips after protection with drug F (89.89). 49±16.00)% decreased to (46.21±14.23)% (P<0.05); (3) After sensitization of guinea pigs protected with drug F, the lung function of the guinea pigs showed a trend of improvement compared with the control group. Conclusion: Drug F can antagonize the contraction response of HA and PAF in lung tissue of guinea pigs in vitro, and it can improve the lung function of sensitized guinea pigs. It can be used for the treatment of bronchial asthma.
