目的研究木贼正丁醇提取物对大鼠动脉粥样硬化早期主动脉平滑肌细胞凋亡及Fas、FasL表达的影响。方法选用雄性SD大鼠,随机分为正常对照、病理模型、阳性药及木贼提取物治疗组。复制食饵性AS早期模型,用木贼提取物预防性给药,以吉非罗齐为阳性药对照。实验9周,流式细胞术定量检测主动脉平滑肌细胞凋亡率及Fas、FasL的表达,光镜下观察主动脉形态学变化。结果木贼正丁醇提取物治疗组平滑肌细胞的凋亡率明显高于模型组(P<0.01),Fas、FasL表达均高于模型组(P<0.01及P<0.05),主动脉损伤程度减轻。结论木贼正丁醇提取物可通过调控AS早期主动脉平滑肌细胞Fas、FasL基因表达,促进平滑肌细胞凋亡,阻断AS进展。
Objective To investigate the effects of n-butanol extracted from Equisetum on the apoptosis of aortic smooth muscle cells and the expression of Fas and FasL in early atherosclerotic rats. Methods Male Sprague-Dawley rats were randomly divided into normal control, pathological model, positive drug and quiks extract treatment group. The precocious AS model was replicated and prophylactically administered with Equisetum extract, with gemfibrozil as the positive control. At the end of the experiment, apoptosis rate of aortic smooth muscle cells and expression of Fas and FasL were detected by flow cytometry quantitatively. Morphological changes of aorta were observed under light microscope. RESULTS: The apoptosis rate of smooth muscle cells in the butanol extract group was significantly higher than that in the model group (P<0.01). The expression of Fas and FasL was higher than that in the model group (P<0.01 and P<0.05). Reduce. Conclusion The n-butanol extract of E. chinense can promote the expression of Fas and FasL genes in early aortic smooth muscle cells of AS, promote the apoptosis of smooth muscle cells and block the progression of AS.
