目的观察不同p H值的赫氏干燥培养基对鼠疫菌生长的影响,从中筛选鼠疫菌生长的最适p H值。方法同一批号的赫氏干燥培养基分别加入到p H值为6.2、6.6、7.0、7.4、7.8、8.2的蒸馏水溶液中,121℃30 min高压灭菌后倾注平皿。通过进行鼠疫菌强毒141标准株和鼠疫菌弱毒EV 76巴黎株菌落计数实验,观察不同p H值的赫氏干燥培养基对鼠疫菌生长的差异。结果不同p H值的赫氏干燥培养基对鼠疫菌的生长均有影响且其差异有统计学意义(EV76χ2=190.804,P<0.05;141χ2=151.742,P<0.05)。EV76株鼠疫菌和141株鼠疫菌在p H值7.0的赫氏干燥培养基中活菌数值最大。结论赫氏干燥培养基对鼠疫菌生长的最适p H值为7.0。
Objective To observe the effects of different pH values of Herzogorsk ’s dry medium on the growth of Y. pestis and to select the optimum p H value for the growth of Y. pestis. Methods The same batch of Herziation drying medium were added to the pH value of 6.2,6.6,7.0,7.4,7.8,8.2 distilled water solution, 121 ℃ 30 min autoclaved after pouring plate. Through the experiments of Yersinia pestis 141 and Yersinia pestis EV 76 Paris strains, the difference of growth of Yersinia pestis between different p H value Hessian drying medium was observed. Results Heptanen ’s drying medium with different p H had an impact on the growth of Y. pestis and the difference was statistically significant (EV76χ2 = 190.804, P <0.05; 141χ2 = 151.742, P <0.05). Yersinia pestis EV71 strains and Yersinia pestis 141 strains had the highest viable count in Hachette drying medium with p H of 7.0. Conclusion The optimum p H value of Herzogam drying medium for the growth of Y. pestis is 7.0.
