为了证实人脐静脉内皮细胞是否具有合成一氧化氮的能力,用NADPH-黄递酶组织化学法和原位杂交技术,对体外培养的人脐静脉内皮细胞表达一氧化氮合酶进行了观察。结果显示,体外培养人脐静脉内皮细胞的NADPH-黄递酶组织化学的阳性百分率为62%,诱生型一氧化氮合酶的原位杂交的阳性百分率为74%。阳性反应产物分布在核周及细胞突起中。本研究首次证明体外培养的人脐静脉内皮细胞能表达诱生型一氧化氮合酶,即人脐静脉内皮细胞在体外培养或受刺激情况下,具有合成一氧化氮的能力。这可能是无神经支配的人脐静脉的一种重要调节因素。
In order to confirm whether human umbilical vein endothelial cells have the ability to synthesize nitric oxide, the expression of nitric oxide synthase in cultured human umbilical vein endothelial cells was observed by NADPH-diaphorase histochemistry and in situ hybridization. The results showed that the positive percentage of NADPH-diaphorase histochemistry in human umbilical vein endothelial cells cultured in vitro was 62%, and the positive percentage of in situ hybridization of inducible nitric oxide synthase was 74%. Positive reaction products are distributed in the perinucleus and cell processes. This study demonstrated for the first time that human umbilical vein endothelial cells cultured in vitro can express inducible nitric oxide synthase, that is, human umbilical vein endothelial cells have the ability to synthesize nitric oxide in vitro cultured or stimulated. This may be an important mediator of the umbilical vein in a non-innervated human.
