目的研究三氯乙烯对大鼠肝细胞凋亡、c-myc和p53表达的作用。方法选用雄性SD大鼠,每组5只,用1 500和3 000 mg/kg的三氯乙烯经口灌胃染毒。用流式细胞术研究细胞凋亡和细胞增殖,用RT-PCR检测c-myc和p53表达。结果雄性SD大鼠经口染毒1 500和3 000 mg/kg的三氯乙烯,可引起肝细胞凋亡和增殖指数下降,肝细胞c-myc mRNA和p53 mRNA较对照组显著增高(P<0.01)。结论三氯乙烯在1 500和3 000 mg/kg的剂量条件下,能明显诱导大鼠肝细胞凋亡并抑制增殖,使大鼠肝细胞c-myc和p53的表达增强。
Objective To study the effects of trichlorethylene on hepatic cell apoptosis, expression of c-myc and p53 in rats. Methods Male Sprague-Dawley rats were randomly divided into five groups. The rats were orally administered with trichlorethylene at 1 500 and 3 000 mg / kg. Flow cytometry was used to study apoptosis and cell proliferation, and c-myc and p53 were detected by RT-PCR. Results The oral administration of trichlorethylene at 1 500 and 3 000 mg / kg in male SD rats resulted in the decrease of hepatocyte apoptosis and proliferation index and the increase of c-myc mRNA and p53 mRNA in hepatocytes compared with the control group (P < 0.01). Conclusion Trichlorethylene at a dose of 1 500 and 3 000 mg / kg significantly induced rat hepatocyte apoptosis and inhibited proliferation, resulting in increased expression of c-myc and p53 in rat hepatocytes.
