目的 用组织化学法,通过观察NADPH-黄递酶的活性了解一氧化氮合酶在豚鼠耳蜗内分布。方法 经4%多聚甲醛心脏灌注固定后,取出耳蜗,经3%依地酸脱钙后,作厚10μm冰冻切片,用辅酶Ⅱ孵育液在37℃条件下孵育1小时。结果 发现在耳蜗内、外毛细胞底部与耳蜗神经末梢接头处及毛细血管球内皮细胞有明显NADPH-黄递酶活性,此外在内、外柱细胞、支持细胞、血管纹及螺旋神经节细胞也有NADPH-黄递酶活性反应。结论 NO在维持耳蜗正常神经传导及毛细血管张力中起着重要作用。
Objective To investigate the distribution of nitric oxide synthase in cochlea of guinea pigs by histochemical method and observe the activity of NADPH-diaphorase. Methods After perfused with 4% paraformaldehyde, the cochleas were removed and decalcified with 3% edetate to make 10μm thick frozen sections. The cells were incubated with coenzyme Ⅱ for 1 hour at 37 ℃. The results showed that in the cochlea, the bottom of the outer hair cells and the cochlear nerve endings joints and capillary endothelial cells have obvious NADPH-diaphorase activity, in addition to the outer column cells, supporting cells, stria vascularis and spiral ganglion cells also NADPH-diaphorase activity response. Conclusion NO plays an important role in maintaining normal cochlear nerve conduction and capillary tone.
