目的以各种应激状态在线虫上模拟应激疲劳综合征(chronic fatigue syndrome,CFS),评估补肾复方对成年线虫应激生存状态及整体寿命的影响。方法在高、中、低浓度(500、250、100 mg/L)补肾复方干预下,观察成年线虫对体外环境的热应激(35℃)、氧化应激(250μg/mL)及体内毒性蛋白应激(Aβ1-42转基因突变株CL4176)的耐受能力和平均存活时间,评估补肾复方对正常状态下线虫平均生存时间(25℃)、20s运动距离、30 s咽泵运动频次及生殖能力的作用效果。结果与对照组比较,补肾复方干预各组均能增加线虫热应激存活时间(P<0.01);高、中浓度干预组可增加线虫氧化应激存活时间,减轻蛋白毒性作用,延长线虫生存时间(P<0.01,P<0.05);高浓度干预组亦能增加线虫的运动距离和咽泵运动频次(P<0.01)。各组间生殖能力比较,差异无统计学意义(P>0.05)。结论补肾复方能增强线虫对内外环境压力的应激能力,延长线虫寿命的同时不干扰线虫正常生殖,提高生命质量,为补肾复方在防治CFS方面的进一步机制探索及药理研究奠定基础。
Objective To simulate the stress response of nematodes on the nematodes of chronic fatigue syndrome (CFS) to evaluate the effects of the prescription for strengthening kidney on the stress survival status and overall life expectancy of adult nematodes. Methods The effects of adult nematodes on heat stress (35 ℃), oxidative stress (250μg / mL) and toxic protein in vivo were observed under high, medium and low doses of 500, 250 and 100 mg / Stress tolerance (Aβ1-42 transgenic mutant CL4176) and mean survival time, evaluate the effect of Bushen recipe on the mean survival time (25 ℃), movement distance of 20s, frequency of 30s pharyngeal pump movement and reproductive capacity Effect. Results Compared with the control group, all the interventions of Bushen Compound could increase the survival time of heat stress of nematode (P <0.01). The intervention group of high and medium concentration could increase the survival time of the oxidative stress of C. elegans, alleviate the toxicity of protein, prolong the survival time of nematode (P <0.01, P <0.05). The high concentration intervention group also increased the movement distance of nematodes and the frequency of pharyngeal pump movement (P <0.01). There was no significant difference in fertility among groups (P> 0.05). Conclusion Bushen Compound can enhance the stress of nematode on the environmental pressure inside and outside, prolong the life of nematodes without interfering with the normal reproduction of nematode and improve the quality of life, and lay a foundation for the further mechanism exploration and pharmacological research of Bushen Compound in the prevention and treatment of CFS.
