AVP主要合成于下丘脑的室旁核和视上核。正常情况下,每毫升血浆AVP含量在10pg以下,近年来的研究表明,疼痛、手术、应激等强刺激可引起AVP大量释放,血浆中的AVP含量可达100pg以上。由于AVP可使血管平滑肌收缩,并具CRF样作用,可促进垂体释放ACTH,因此被认为是一种应激激素。而下丘脑.垂体-肾上腺皮质轴被认为是经典的应激系统。本工作旨在通过去除应激轴上的肾上腺这一环节,来验证AVP是否直接参与应激活动。我们应用放射免疫测定法(RlA)测定了去肾上腺大鼠游泳时血浆、脑垂体及下丘脑内ir-AVP含量的变化。观察了游泳1min、10min、20min、30min和没顶时,血浆、脑垂体、下丘脑内的ir-AVP含量的变化。结果表明,冷水游泳时,大鼠下丘脑ir-AVP含量增高,1min、20min时增加显著(p<0.005);脑垂体内ir-AVP含量减少(p<0.01):血浆ir-AVP含量在1~30min时有升高趋势,但无统计学意义,没顶时ir-AVP含量显著增加(p<0.05)。以上结果表明,冷水游泳对去肾上腺大鼠下丘脑、脑垂体和血浆中AVP含量有明显影响,提示AVP不仅通过激活下丘脑-垂体-肾上腺皮质系统参与应激活动,而且AVP可能直接参与冷水游泳应激活动的调节
AVP is mainly synthesized in the paraventricular nucleus and supraoptic nucleus of the hypothalamus. Under normal circumstances, the plasma AVP levels per milliliter below 10pg, recent studies have shown that pain, surgery, stress and other strong stimulation can cause a large number of AVP release, the plasma AVP content of up to 100pg or more. AVP is thought to be a stress hormone because it attenuates vascular smooth muscle and has CRF-like effects that promote the release of ACTH from the pituitary. The hypothalamus, pituitary-adrenal axis is considered a classic stress system. This work aims to verify whether AVP is directly involved in stress by removing the adrenal part of the stress axis. We used radioimmunoassay (RIA) to measure the changes of ir-AVP in plasma, pituitary and hypothalamus of adrenal rats during swimming. The changes of the content of ir-AVP in plasma, pituitary and hypothalamus were observed after 1 min, 10 min, 20 min, 30 min swimming and no swimming. The results showed that the level of ir-AVP in the hypothalamus of rats increased when swimming in cold water for 1min and 20min (p <0.005), and the content of ir-AVP in pituitary gland decreased (p <0.01) ~ 30min, but there was no statistical significance. The content of ir-AVP increased significantly (p <0.05) at the time of no-top. The above results show that cold water swimming has significant effect on the content of AVP in hypothalamus, pituitary and plasma of adrenal rats, suggesting that AVP not only participates in the stress activity by activating the hypothalamus-pituitary-adrenal cortex system, but also AVP may directly participate in cold water swimming Regulation of stressful activity
