目的:观察心脉康胶囊对冠脉结扎模型犬急性心肌缺血的干预作用。方法:健康杂种犬25条,雌雄兼用,随机分为模型组(生理盐水)、地奥心血康对照组(40 mg/kg)及心脉康低(1.55 g/kg)、中(3.10 g/kg)、高(6.20g/kg)剂量组,每组5只。结扎冠脉左前降支复制心肌缺血模型,灌胃给药后分别抽血做血气分析与心肌酶检测并记录给药前后各时间点的心外膜电图,同时监测血压及心率,于240 min后,取心肌组织NBT染色,测定梗死的范围(梗死区/心脏质量×100%)。结果:(1)心脉康胶囊各剂量在给药后120 min能显著增加犬的冠脉血流量(CBF),降低犬冠脉阻力(CVR)(P<0.05或P<0.01);(2)心脉康胶囊高、中剂量能显著降低犬的心肌耗氧量(MVO2)(P<0.05或P<0.01);(3)心脉康胶囊高、中剂量能显著减小犬的心肌梗死范围(P<0.01);(4)心脉康胶囊能显著降低犬血清乳酸脱氢酶(LDH)及肌酸磷酸激酶(CPK)的水平(P<0.05或P<0.01)。结论:心脉康胶囊具有显著保护心肌、抗心肌缺血的作用,可作为防治心肌缺血的有效药物。
Objective: To observe the intervention effect of Xin Mai Kang capsule on acute myocardial ischemia in canine model of coronary artery ligation. METHODS: Twenty-five healthy mongrel dogs, both male and female, were randomly divided into model group (normal saline), Dioxinxuekang control group (40 mg/kg), Xinmaikang low (1.55 g/kg), and medium (3.10 g/kg). High (6.20 g/kg) dose groups, 5 per group. The left anterior descending coronary artery was ligated to replicate a myocardial ischemia model. After intragastric administration, blood was collected for blood gas analysis and myocardial enzyme detection, and epicardial electrograms at various time points before and after administration were recorded. Blood pressure and heart rate were monitored at the same time. After min, myocardial tissue was stained with NBT to determine the extent of infarct (infarct zone/cardiac mass × 100%). Results: (1) Each dose of Xinmaikang capsule can significantly increase coronary blood flow (CBF) and decrease canine coronary resistance (CVR) in dogs 120 min after administration (P<0.05 or P<0.01); (2) ) Xinmaikang capsules high and medium dose can significantly reduce the myocardial oxygen consumption (MVO2) (P <0.05 or P <0.01); (3) Xinmaikang capsule high and medium dose can significantly reduce the myocardial infarction in dogs Scope (P<0.01); (4) Xinmaikang capsule can significantly reduce the serum lactate dehydrogenase (LDH) and creatine phosphokinase (CPK) levels in dogs (P<0.05 or P<0.01). Conclusion: Xinmaikang capsule has the effect of protecting myocardium and preventing myocardial ischemia. It can be used as an effective drug for preventing and treating myocardial ischemia.
