目的探讨骨髓间充质干细胞对胃溃疡愈合质量的影响。方法大鼠乙酸胃溃疡模型复制3 d后随机分为3组。模型组:只做剖腹手术;磷酸盐缓冲液组:剖腹手术后,在溃疡周围胃壁浆膜下注入磷酸盐缓冲液;干细胞组:剖腹手术后,在溃疡周围胃壁浆膜下注入干细胞。25 d后用组织学方法检测再生胃黏膜扩张腺体数和胃黏液,用免疫组织化学方法检测环加氧酶-2的表达,用酶联免疫测定方法检测胃黏膜前列腺素E2含量,用血流仪检测再生黏膜部位血流量。结果干细胞组的扩张腺体数低于模型组(P<0.01)和磷酸盐缓冲液组(P<0.01),干细胞组的环加氧酶-2和胃黏液积分光密度高于模型组(P<0.01)和磷酸盐缓冲液组(P<0.01),干细胞组的胃黏膜前列腺素E2含量和黏膜血流量高于模型组(P<0.01)和磷酸盐缓冲液组(P<0.01)。结论骨髓间充质干细胞植入溃疡周围能够提高胃溃疡愈合质量。
Objective To investigate the effect of bone marrow mesenchymal stem cells on healing quality of gastric ulcer. Methods Rat gastric ulcer model was randomly divided into 3 groups after 3 days of replication. Model group: laparotomy only; Phosphate buffer group: After laparotomy, phosphate buffer solution was infused into the submerged gastric wall under the laparotomy; Stem cell group: After laparotomy, stem cells were injected into the submerged gastric wall under the ulcer. Twenty-five days later, the number of gastric mucosal dilatation glands and gastric mucosa were detected by histological method. The expression of cyclooxygenase-2 was detected by immunohistochemical method. The content of prostaglandin E2 in gastric mucosa was detected by enzyme linked immunosorbent assay Flow meter detection of mucosal regeneration of blood flow. Results The numbers of dilated glands in stem cells group were lower than those in model group (P <0.01) and phosphate buffered saline group (P <0.01). The optical density of cyclooxygenase-2 and gastric mucus in stem cell group was higher than that in model group (P <0.01) and phosphate buffer group (P <0.01). The content of prostaglandin E2 and mucosal blood flow in the stem cell group were higher than those in the model group and phosphate buffer group (P <0.01). Conclusion Bone marrow mesenchymal stem cells can improve the quality of gastric ulcer healing when implanted around the ulcer.
