目的:探讨加味温胆汤对抑郁模型大鼠海马神经元再生变化的影响。方法:以孤养结合慢性不可预见性应激抑郁模型大鼠为研究对象,在实验第7、14、21、28 d,采用免疫荧光双标技术检测各组大鼠海马神经元增殖与分化的再生变化情况。结果:在抑郁形成过程中,大鼠海马神经元NeuN/BrdU、β-tubulinⅢ/BrdU双标细胞数逐渐降低,至21、28 d时模型组NeuN/BrdU、β-tubulinⅢ/BrdU双标阳性细胞数明显减少,经中、西药治疗后,大鼠海马神经元NeuN/BrdU、β-tubulinⅢ/BrdU双标阳性细胞数下降趋势得到抑制,与模型组比较增加明显(P<0.05或P<0.01)。结论:抑郁形成过程中海马神经元增殖与分化减少的再生障碍渐次加重,加味温胆汤可逆转此现象。
Objective: To investigate the effect of Jiawei Wendan decoction on neuronal regeneration in hippocampus of depression model rats. Methods: The isolated and cultured chronic unpredictable stress depression model rats as the research object, on the 7th, 14th, 21st and 28th day, the immunofluorescence double labeling technique was used to detect the proliferation and differentiation of hippocampal neurons in each group Regeneration changes. Results: During the process of depression, the numbers of NeuN / BrdU and β-tubulinⅢ / BrdU double-labeled neurons in hippocampus of rats gradually decreased. At 21 and 28 days, the positive cells of NeuN / BrdU and β-tubulinⅢ / (P <0.05 or P <0.01). Compared with the model group, the numbers of positive cells of NeuN / BrdU and β-tubulinⅢ / BrdU in the hippocampus decreased significantly . CONCLUSION: The regeneration disorder of hippocampal neurons with decreasing proliferation and differentiation during the formation of depression gradually increases. Modified Wendan Decoction can reverse this phenomenon.
