目的:测定马鹿茸不同分子量肽段中水解氨基酸和游离氨基酸的含量及构成。方法:采用柱前衍生化,氨基酸自动分析仪对马鹿茸肽段中氨基酸类成分进行含量测定。色谱柱为氨基酸分析专用ODS柱,柱温:27℃;检测波长360 nm;流动相总流速1.2 mL·min~(-1)。结果:马鹿茸肽段中氨基酸类成分线性关系良好,r均大于0.999;精密度、稳定性、重复性试验的RSD均小于3%;加样回收率在96.6%~104.7%之间,RSD值均小于2.4%。不同分子量肽段的氨基酸含量及构成有一定差异。50~100 k分子量段的氨基酸含量最高。结论:该方法简便可行、重复性好、准确度高,可用于马鹿茸药材多肽中水解氨基酸及游离氨基酸的含量测定。
Objective: To determine the content and composition of hydrolytic amino acids and free amino acids in the different molecular weight peptides of the antler. Methods: The contents of amino acids in the antler antler peptide were determined by precolumn derivatization and automatic amino acid analyzer. The chromatographic column was an ODS column for amino acid analysis. The column temperature was 27 ℃, the detection wavelength was 360 nm, and the total mobile phase flow rate was 1.2 mL · min ~ (-1). Results: The amino acid composition of the antler antler peptide had a good linear relationship (r> 0.999). The RSDs of precision, stability and reproducibility were all less than 3%. The recoveries were between 96.6% and 104.7% Less than 2.4%. There were some differences in amino acid content and composition between different molecular weight peptides. 50 ~ 100 k molecular weight amino acid content of the highest. Conclusion: The method is simple, feasible, reproducible and accurate. It can be used to determine the content of hydrolyzed amino acids and free amino acids in the polypeptide.
