目的研究山核桃叶总黄酮诱导人多发性骨髓瘤(U266)细胞株凋亡的作用及其作用机制。方法体外培养U266细胞,CCK-8比色法检测细胞增殖活性,流式细胞术定量分析细胞凋亡程度,Western Blot分析不同浓度总黄酮对U266细胞蛋白激酶B(Akt)和p-Akt蛋白表达的影响。结果山核桃叶总黄酮以浓度依赖性方式显著诱导U266细胞株凋亡并抑制p-Akt蛋白的表达。结论山核桃叶总黄酮诱导U266细胞凋亡的作用与其抑制Akt磷酸化相关。
Objective To study the effect and mechanism of total flavonoids from Pecan Lathyrus leaf on apoptosis in human multiple myeloma (U266) cell line. Methods U266 cells were cultured in vitro. The proliferation of U266 cells was detected by CCK-8 colorimetric assay. The degree of apoptosis was quantified by flow cytometry. The protein expression of Akt and p-Akt in U266 cells was analyzed by Western Blot Impact. Results The total flavonoids of pecan leaves significantly induced apoptosis of U266 cells and inhibited the expression of p-Akt protein in a concentration-dependent manner. Conclusion The effect of total flavonoids from Hickory Nutcracker leaves on the apoptosis of U266 cells is related to the inhibition of Akt phosphorylation.
