目的研究内脏脂肪素在糖尿病小鼠心肌组织中的表达。方法将昆明小鼠随机分为对照(NC)组和糖尿病(DM)组,DM组采用STZ造模。每周检测一次体重、血糖。利用透视电子显微术分析心肌超微结构变化,免疫印迹法观察内脏脂肪素在心肌组织中表达情况。结果 4周后,与NC组相比,DM组体重和血糖显著升高(P<0.05)。DM组心肌细胞超微结构显示染色质凝集、闰盘各带连接明显扩张、线粒体嵴断裂空变、肌丝溶解被大量空泡填充。与NC组相比,DM组心肌组织内脏脂肪素蛋白表达水平增加(P<0.05)。结论高血糖能够破坏心肌细胞,在糖尿病小鼠心肌组织中内脏脂肪素蛋白表达水平增加,可能是一种代偿反应,对心肌起到一定的保护作用。
Objective To study the expression of visfatin in myocardium of diabetic mice. Methods Kunming mice were randomly divided into control (NC) group and diabetes mellitus (DM) group. STZ model was used in DM group. Weekly test weight, blood sugar. The changes of myocardial ultrastructure were analyzed by fluoroscopy, and the expression of visfatin in myocardial tissue was observed by immunoblotting. Results After 4 weeks, body weight and blood glucose in DM group were significantly higher than those in NC group (P <0.05). Myocardial ultrastructure of DM group showed chromatin condensation, intercalated disc with a significant expansion of each link, mitochondrial cristae fracture change, myofilament was filled with a large number of vacuoles. Compared with NC group, the expression of visfatin in DM group increased (P <0.05). Conclusion Hyperglycemia can damage cardiomyocytes and increase the expression of visfatin in the myocardium of diabetic mice, which may be a compensatory response and play a protective role in the myocardium.
