目的:探讨特异性多肽P161与顺铂(CDDP)联合应用时乳腺癌细胞株MA-782增殖及凋亡的影响。方法:应用四甲基偶氮唑比色法(MTT)测定P161、CDDP单用及联合应用对MA-782细胞增殖的抑制,光学显微镜观察细胞形态学改变,荧光显微镜观察经Hoechst33342染色后细胞凋亡形态改变,流式细胞术检测细胞凋亡率。结果:MTT显示,与单独用药组相比,联合用药组细胞增殖活力显著降低,呈剂量依赖性,且与CDDP组比较,差异有统计学意义(P<0.05);倒置显微镜观察,与单独给药组相比,联合用药组细胞形态改变愈加明显;Hoechst33342染色显示,与单独用药组相比,联合用药组凋亡形态特征更为显著,染色质凝集现象增多。流式细胞术检测显示,与CDDP组相比,联合给药组细胞早期凋亡率显著升高,其凋亡率由(30.03±0.93)%增至(61.67±0.81)%(P<0.01)。结论:多肽P161联合CDDP可增加乳腺癌细胞株对CDDP的敏感性,此作用可能与促进细胞早期凋亡有关。
Objective: To investigate the effects of specific polypeptide P161 and cisplatin (CDDP) on proliferation and apoptosis of breast cancer cell line MA-782. Methods: The inhibitory effect of P161 and CDDP alone or in combination on the proliferation of MA-782 cells was determined by MTT assay. The morphological changes of the cells were observed by light microscopy. The apoptosis of the cells was observed by fluorescence microscopy after Hoechst33342 staining Morphological changes were observed by flow cytometry apoptosis rate. Results: MTT showed that the cell proliferation activity of the combination group was significantly lower than that of the drug-alone group in a dose-dependent manner, and the difference was statistically significant (P <0.05) compared with the CDDP group; inverted microscopy, Hoechst33342 staining showed that compared with the drug alone group, the morphological changes of apoptosis in the combination group were more obvious and the chromatin agglutination phenomenon increased. Flow cytometry showed that compared with CDDP group, the apoptosis rate of the combination group increased significantly from (30.03 ± 0.93)% to (61.67 ± 0.81)% (P <0.01) . Conclusion: Polypeptide P161 combined with CDDP can increase the sensitivity of breast cancer cell lines to CDDP, and this effect may be related to the promotion of early apoptosis of cells.
