目的:研究白术茯苓汤不同配比水煎液中多糖的含量;方法:水提醇沉法分离出粗多糖,采用蒽酮-硫酸法制得有色糖醛衍生物,用分光光度法进行含量测定。结果:加入0.3%的硫酸蒽酮溶液4mL,在水浴中加热10min,冷却10min时复方多糖含量测定条件最优。显示627nm处有最大吸收峰,线性关系良好(r=0.9997),平均回收率为97.3%。结论:白术单煎多糖提取的含量最高,茯苓所占比例越大,含量越低。该方法快速准确,重现性好。
OBJECTIVE: To study the content of polysaccharides in the decoction of different doses of Atractylodes Tuckahoe Decoction. Methods: The crude polysaccharides were separated by water-alcohol precipitation method, and the derivatives were determined by anthrone-sulfuric acid method. The content of polysaccharides was determined by spectrophotometry. Results: 4 mL of 0.3% sulfuric acid anthrone solution was added and heated in a water bath for 10 min. The optimum conditions for the determination of polysaccharide content were obtained after cooling for 10 min. The peak at 627 nm showed a good linear relationship (r = 0.9997) with an average recovery of 97.3%. Conclusion: Atractylodes macrocephala polysaccharide is the highest content of extraction, the larger the proportion of Poria, the lower the content. The method is fast and accurate and reproducible.
