[目的 ]从分子生物学角度进一步证实我们发现的 1例 HIV感染者为国内首例 HIV- 2型病毒携带者。 [方法 ]从 Genebank查询已知的 HIV- 2核酸序列 ,通过序列比较以及酶切位点分析查找 HIV- 2基因组中保守区 ,应用套式PCR扩增出 HIV- 2基因组 L TR- GAG区的~ 330 bp的核酸片段 ,并经限制性酶切位点和序列分析证实该扩增片段为HIV- 2特异性片段。 [结果 ]实验表明 ,扩增片段为 HIV- 2特异性片段 ,序列分析也表明感染者体内病毒为 HIV- 2。 [结论 ]从遗传学上进一步支持该感染者为 HIV- 2携带者。
[Objective] To further confirm from the perspective of molecular biology that one case of HIV infection we found was the first case of HIV-2 virus in China. [Method] The known HIV-2 nucleic acid sequence was searched from Genebank, and the conserved region of HIV-2 genome was searched by sequence comparison and restriction enzyme analysis. The nested PCR was used to amplify the HIV-2 LTR-GAG region ~ 330 bp nucleic acid fragment. The restriction fragment and sequence analysis confirmed that the amplified fragment was HIV-2 specific fragment. [Result] The experiment showed that the amplified fragment was HIV-2 specific fragment, and the sequence analysis also showed that the virus in the infected person was HIV-2. [Conclusion] The genetics further supports this HIV-2 carrier.
