目的:研究通络救脑注射液中三七总皂苷、栀子苷组分配伍对拟缺血损伤大鼠脑微血管内皮细胞氧化应激反应的药效特征。方法:采用原代培养大鼠脑微血管内皮细胞,氧糖剥夺法制备拟缺血损伤模型,三七总皂苷、栀子苷以及通络救脑注射液干预后,采用CCK8法测定各组微血管内皮细胞活性的变化,生化法检测丙二醛(Malonic dialdehyde,MDA)含量、超氧化物歧化酶(Superoxidase dismutase,SOD)活性。结果:3种药物均可有效提高拟缺血损伤内皮细胞活性,其中栀子苷在降低脂质过氧化的产物MDA含量上优于三七总皂苷,而三七总皂苷在提高氧自由基清除酶SOD活性上优于栀子苷,通络救脑注射液均优于两有效组分。结论:栀子苷的抗氧化作用可能与抑制自由基的脂质过氧化反应途径有关,三七总皂苷可能与增强自由基的清除有关,通络救脑注射液整体上明显优于2个组分,显示了中药组方配伍发挥整合调节的治疗优势。
OBJECTIVE: To study the pharmacodynamic characteristics of the combination of Panax notoginseng saponins and geniposide in Tongluojiannao injection on oxidative stress in brain microvascular endothelial cells of rats with ischemic injury. Methods: The model of ischemic injury was prepared by primary culture of rat brain microvascular endothelial cells and the model of ischemic injury was induced by OGT. After the intervention of Panax Notoginseng Saponins, Geniposide and Tongluo Jiu Nao Injection, CCK8 method was used to detect the microvascular endothelium The changes of cell viability were measured. The levels of malonic dialdehyde (MDA) and superoxide dismutase (SOD) were detected by biochemical methods. Results: All the three drugs could effectively improve the activity of endothelial cells in ischemic injury. Geniposide was superior to Panax notoginseng saponins in reducing the MDA content of products of lipid peroxidation, while the total saponins of Panax notoginseng in improving the clearance of oxygen free radicals Enzymatic activity of SOD is superior to geniposide, Tongluo brain injection are better than the two active ingredients. Conclusion: The antioxidative effect of geniposide may be related to the inhibition of free radical lipid peroxidation pathway. The total saponin of Panax notoginseng may be related to the enhancement of free radical scavenging. Tongluojiannao injection is obviously better than the other two groups Points, shows the traditional Chinese medicine group to play a combination of regulation and treatment advantages.
