目的:研究有氧运动和膳食控制对2型糖尿病大鼠骨骼肌胰岛素受体亲和力的影响。方法:62只雄性大鼠,随机分为正常对照组(C组),DM对照组(DM)、DM+运动锻炼组(DME)、DM+膳食控制组(DMD)、DM+运动锻炼+膳食控制组(DMED),用受体放射性配基结合分析法(RBA)测定各组大鼠骨骼肌胰岛素受体亲和力。结果表明:1)与正常大鼠比较,糖尿病大鼠骨骼肌胰岛素高、低亲和力受体亲和力常数KD1、KD2都有非常显著性增加(P<0.01),受体结合容量RT1也有非常显著性增加(P<0.001),RT2显著性增加(P<0.05);2)与糖尿病对照组比较,糖尿病运动组KD1和RT2没有显著性变化(P>0.05),KD2有非常显著性下降(P<0.001),RT1有显著性下降(P<0.05);3)与糖尿病对照组比较,膳食控制组KD1、KD2、RT1和RT2都没有显著性变化(P>0.05),且运动和膳食控制没有交互作用(P>0.05)。结论:糖尿病大鼠胰岛素受体亲和力出现了显著性下降,有氧运动能够增强糖尿病大鼠胰岛素受体结合力,但膳食控制对其作用不明显,且与运动没有交互作用。说明胰岛素受体在糖尿病的发病机制及运动干预过程中都起到了重要的作用。
Objective: To study the effects of aerobic exercise and dietary control on the insulin receptor affinity of skeletal muscle in type 2 diabetic rats. Methods: Sixty two male rats were randomly divided into normal control group (DM group), DM control group (DM), DM + exercise group (DME), DM + DMD group, DM + exercise group + DMED) were used to detect the insulin receptor affinity of skeletal muscle of rats in each group by radioimmunoassay (RBA). The results showed that: 1) compared with normal rats, the KD1 and KD2 affinity constants of skeletal muscle with high insulin and low affinity of skeletal muscle of diabetic rats increased significantly (P <0.01), and the receptor binding capacity of RT1 also had a very significant increase (P <0.001) and RT2 (P <0.05) .2) Compared with diabetic control group, there was no significant change in KD1 and RT2 in diabetic exercise group (P> 0.05), KD2 was significantly decreased (P <0.001) (P <0.05) .3) Compared with diabetic control group, there was no significant change in KD1, KD2, RT1 and RT2 in diet control group (P> 0.05), and there was no interaction between exercise and diet control (P> 0.05). CONCLUSION: The insulin receptor affinity of diabetic rats shows a significant decrease. Aerobic exercise can increase the insulin receptor binding ability of diabetic rats. However, dietary control has no obvious effect on the insulin receptor binding and has no interaction with exercise. Insulin receptors in the pathogenesis of diabetes and exercise intervention have played an important role.
