目的探讨外源性肾上腺髓质素(ADM)对肾脏机械性损伤早期肾小管评分的影响。方法选健康成年普通级Wistar大鼠104只,随机分为4组:正常对照组8只,创伤组32只,创伤前注射ADM组32只,创伤后注射ADM组32只。后3组动物分别于创伤后1、6、12和24 h各处死8只,采用自由落体打击仪直接打击大鼠脊肋区制作肾机械性损伤模型。两给药组分别于创伤前、后10 min腹腔注射ADM(0.1 nmol/kg)。3组肾损伤大鼠平均分为4批,分别于创伤后1、6、12和24 h迅速解剖动物提取下肾脏标本,10%甲醛固定。HE染色观察创伤后肾组织的基本病理改变。结果单纯创伤组于创伤早期(1、6 h)肾小管评分明显升高(P<0.05),急性创伤后期(12、24 h)上述指标逐渐降低,接近正常对照组(P>0.05)。外源性ADM可降低肾小管评分。结论肾脏损伤期间给予外源性ADM对肾组织均有明显的保护作用,其作用强弱与给药时间关系密切。
Objective To investigate the effect of exogenous adrenomedullin (ADM) on early renal tubule score of renal mechanical injury. Methods A total of 104 healthy adult Wistar rats were randomly divided into 4 groups: normal control group (n = 8), trauma group (n = 32), pretraumatic ADM group (n = 32) and traumatic ADM group (n = 32). After three groups of animals were sacrificed 1, 6, 12 and 24 h after the wounds were killed eight, the use of free-fall bashing instrument directly against the rat’s rib cage manufacturing mechanical injury model of kidney. Two administration groups were injected with ADM (0.1 nmol / kg) intraperitoneally 10 min before and after trauma. The rats in the 3 groups were divided into 4 batches on average. The kidneys were rapidly dissected from animals at 1, 6, 12 and 24 h after trauma, respectively, and fixed with 10% formaldehyde. HE staining was used to observe the basic pathological changes of renal tissue after trauma. Results The score of renal tubules in trauma group was significantly higher than that in normal control group (1,6 h) (P <0.05). The index of traumatic injury group (12 and 24 h) was lower than that of normal control group (P> 0.05). Exogenous ADM reduces tubule score. Conclusions Administration of exogenous ADM during renal injury has a significant protective effect on renal tissue, and its effect is closely related to the time of administration.
