目的探讨新生鼠肠损伤时MMP-2 mRNA和MMP-9 mRNA的动态表达,并比较肠损伤程度与肠黏膜MMP-2 mRNA和MMP-9 mRNA关系,为临床预防和治疗NEC提供理论依据。方法1日龄新生鼠随机分实验组腹腔注射5mg/kg EcoliO55:B5内毒素(LPS),对照组注射等量生理盐水,于LPS注射后1h、3h、6h、12h和24h处死分离胃肠道,部分回肠组织用于形态学观察,其余组织用于MMP-2 mRNA和MMP-9 mRNA测定。结果LPS注射后1h、3h、6h、12h和24h回肠组织平均损伤评分分别为1.28±0.62,1.75±0.74,1.98±0.75,2.85±0.41,2.35±0.63,均显著高于对照组(0.12±0.17)(P<0.01),高峰在12h。对照组MMP-2 mRNA和MMP-9 mRNA已有表达,内毒素注射后初期MMP-2 mRNA和MMP-9 mRNA几无表达,随着时间的延续,逐渐上调,在12h达到高峰,至24h反而下降;而MMP-9mR-NA与肠损伤程度呈正相关(P<0.05);但MMP-2 mRNA与肠损伤程度无显著相关性(P>0.05)。结论LPS致新生鼠肠损伤时,回肠组织MMP-9不但与肠黏膜损伤密切相关,可能还参与了肠黏膜屏障的修复机制。
Objective To explore the dynamic expression of MMP-2 mRNA and MMP-9 mRNA in intestinal mucosa of neonatal rats and to compare the relationship between intestinal damage and the expression of MMP-2 mRNA and MMP-9 mRNA in intestinal mucosa, so as to provide a theoretical basis for clinical prevention and treatment of NEC. Methods One-day-old newborn rats were randomly divided into experimental group (5mg / kg EcoliO55: B5), intraperitoneal injection of normal saline, control group were injected with normal saline at 1h, 3h, 6h, 12h and 24h after LPS injection, , Partial ileum tissue for morphological observation, and the remaining tissue for MMP-2 mRNA and MMP-9 mRNA determination. Results The average damage scores of ileal tissue at 1h, 3h, 6h, 12h and 24h after LPS injection were 1.28 ± 0.62,1.75 ± 0.74,1.98 ± 0.75,2.85 ± 0.41,2.35 ± 0.63, which were significantly higher than those in control group (0.12 ± 0.17 ) (P <0.01), peak at 12h. The expression of MMP-2 mRNA and MMP-9 mRNA in the control group was already expressed. After the endotoxin injection, the expression of MMP-2 mRNA and MMP-9 mRNA did not express much at the initial stage, and gradually increased with time, reached the peak at 12h, (P <0.05). However, there was no significant correlation between MMP-2 mRNA and intestinal injury (P> 0.05). Conclusions MMP-9 in ileum is not only associated with intestinal mucosal injury but also involved in the repair mechanism of intestinal mucosal barrier.
