目的:观察不同剂量咖啡酸对白细胞及血小板减少症模型小鼠外周血血小板膜糖蛋白CD62p、CD41及CD61表达水平的影响。方法:采用阿糖胞苷制备白细胞及血小板减少症小鼠模型,同时每天一次灌胃给予低(100mg/kg)、中(150mg/kg)、高(200mg/kg)不同剂量咖啡酸,阴性对照组同法给予等量溶媒0.5%羧甲基纤维素钠,阳性对照组给予重组白细胞介素-11。结果:给药第10天,咖啡酸中剂量及高剂量组血小板膜糖蛋白CD62p的表达水平明显高于正常及模型阴性对照组;各模型组血小板膜糖蛋白CD41及CD61表达水平较正常明显降低;与阴性对照组比较,阳性对照组及咖啡酸各剂量组对CD41及CD61表达无明显影响。结论:咖啡酸中剂量及高剂组能明显升高模型小鼠血小板膜表面的CD62p表达水平,活化血小板,对于启动和扩大血栓形成,从而增强凝血功能具有重要意义。
Objective: To observe the effect of different doses of caffeic acid on the expression of platelet membrane glycoprotein CD62p, CD41 and CD61 in peripheral leukocyte and thrombocytopenic mice. Methods: The white blood cells and thrombocytopenia mouse models were prepared by cytarabine. At the same time, low dose (100mg / kg), middle dose (150mg / kg) and high dose (200mg / kg) The same group given the same amount of vehicle 0.5% sodium carboxymethyl cellulose, positive control group given recombinant interleukin-11. Results: On the 10th day after administration, the expression level of platelet membrane glycoprotein CD62p in Caffeic acid medium and high dose groups was significantly higher than that in normal and model negative control groups. The expression levels of platelet membrane glycoproteins CD41 and CD61 in model group were significantly lower than those in normal control ; Compared with the negative control group, positive control group and each dose of caffeic acid had no significant effect on the expression of CD41 and CD61. CONCLUSION: Caffeic acid medium dose and high dose group can significantly increase the expression level of CD62p on platelet membrane and activate platelet, which is of great significance for the initiation and expansion of thrombosis, thereby enhancing the coagulation function.
