目的鉴定长春地区孕妇感染弓形虫株的基因型。方法采用间接ELISA法检测长春地区840名孕妇血清中抗弓形虫IgG抗体,阳性者提取全血DNA,以弓形虫529bp高度重复序列为靶基因进行PCR扩增,用多基因位点聚合酶链反应-限制性片段长度多态性(PCR-RFLP)分析鉴定PCR阳性标本的基因型。结果 ELISA检测80份标本IgG阳性,其中33份标本同时PCR阳性。对PCR阳性标本进行PCR-RFLP分型鉴定,有8份标本为基因Ⅱ型。结论长春地区孕妇感染的弓形虫主要为基因Ⅱ型。
Objective To identify genotypes of Toxoplasma gondii in pregnant women in Changchun area. Methods Anti-Toxoplasma gondii IgG antibodies were detected in serum of 840 pregnant women in Changchun by indirect ELISA. The positive samples were extracted from whole blood DNA. PCR was performed using the 529bp highly repetitive sequence of Toxoplasma gondii as target gene. Polymerase chain reaction Restriction fragment length polymorphism (PCR-RFLP) analysis identified genotypes of PCR-positive specimens. Results ELISA test 80 samples were positive for IgG, of which 33 were PCR positive at the same time. PCR-RFLP typing PCR positive samples were identified, 8 samples for the genotype Ⅱ. Conclusion Toxoplasma gondii infection in pregnant women in Changchun area is mainly genotype Ⅱ.
