目的探索体外窦房结细胞原代纯化培养方法,观察培养的窦房结细胞生长状况及光电镜形态学特征。方法(1)用差速贴壁和Brdu处理作细胞培养;(2)观察和记录各类型培养细胞的生长状况;(3)HE染色和Heidenhain苏木素染色;(4)透射电镜观察。结果在培养的窦房结细胞中,至少有3种自发性收缩细胞及少量的星状和不规则细胞。收缩性细胞中,最多的是多边形细胞,其次是梭形,最少的是三角形细胞,且多边形细胞收缩频率最低(75.27±1.73/minx±s),细胞染色和电镜观察结果显示:多边形细胞的细胞器相似于原位窦房结组织中的起搏(P)细胞,梭形细胞相似于移行(T)细胞,P细胞核旁较易见到电子致密颗粒和中心粒,也常见细胞核染色体正处于细胞周期的分裂期的前、中期。结论多边形细胞很幼稚,具有不断分裂增埴的能力。
Objective To explore the method of primary purification and culture of extracellular matrix of sinoatrial node cells and to observe the growth of cultured sinoatrial node cells and the morphological characteristics of light microscope. Methods (1) Cell culture was performed by differential adherent and Brdu treatment; (2) Observation and recording of the growth of various types of cultured cells; (3) HE staining and Heidenhain hematoxylin staining; (4) Transmission electron microscope observation. Results In cultured sinoatrial node cells, there were at least 3 spontaneous contractile cells and a few asteroidal and irregular cells. Systolic cells, the most is polygonal cells, followed by the spindle shape, the smallest is the triangular cells, and the lowest polygular contraction frequency (75.27 ± 1.73 / minx ± s), cell staining and electron microscopy showed that: Organelles of polygonal cells are similar to pacing (P) cells in situ sinus node tissue. Fibroblast cells are similar to transitional (T) cells. Electronically dense particles and centrioles are more likely to be seen next to the nucleus of P cells. Nuclei chromosomes are also common Is in the cell cycle of the division of the former and mid-term. Conclusion Polygon cells are naive and have the ability to continuously divide and proliferate.
