嗜酸性集落形成细胞(EO-CFC)存在于正常骨髓中,许多方面与粒-单核集落形成细胞(GM-CFC)有所不同。虽然已知GM-CFC主要属于周围血液淋巴细胞的N细胞亚群,但尚不知EO-CFC是否也属于N细胞。作者取正常人骨髓及周围血液用FicollHypaque 沉降法分离细胞,做软琼脂培养,用抗氰化物过氧化物酶(Cyanide-ri-sistant Peroxidase)活性染色或根据细胞呈致密包裹样生长来鉴定嗜酸性细胞集落。用E花结和EAC花结试验,或用Leu-1和HLA DR单克隆抗体检查膜标志。实验结果如下:嗜酸性细胞集落比粒-单核细胞集落增殖缓慢,培养21天集落数最多,培养28天集落数不再增多,但集落变大,骨髓嗜酸性
Eosinophilic colony forming cells (EO-CFCs) are present in normal bone marrow in many respects distinct from granulocyte-mononuclear colony forming cells (GM-CFCs). Although GM-CFCs are known to belong primarily to the N-cell subset of peripheral blood lymphocytes, it is unknown whether EO-CFCs also belong to N-cells. The authors take normal human bone marrow and peripheral blood by FicollHypaque sedimentation separation of cells, do soft agar culture, anti-cyanide peroxidase (Cyanide-ri-sistant Peroxidase) activity staining or cell-like dense parcel-like growth to identify eosinophilic Cell colonies. E-rosette and EAC flowering assay, or membrane labeling with Leu-1 and HLA DR monoclonal antibodies.
