白三烯B_4为花生四烯酸5-脂氧酶代谢产物,是炎症反应中的重要介质。目前抑制白三烯B_4生物合成的药物尚不多见。本文建立了测定白细胞来源白三烯B_4的反相高效液相色谱法,并初步探讨了几种抗炎药对白三烯B_4生物合成的影响。结果表明阿斯匹林、消炎痛、炎痛喜康、氢化可的松及麝香第一色带对白三烯B_4生物合成几无影响,而氟灭酸可明显抑制白三烯B_4生成,提示它可能是5-脂氧酶抑制剂。
Leukotriene B_4 is arachidonic acid 5-lipoxygenase metabolites, is an important medium in the inflammatory response. The current inhibition of leukotriene B 4 biosynthesis drugs are rare. In this paper, a reversed-phase high performance liquid chromatography (RP-HPLC) was established for the determination of Leukocyte-derived leukotriene B_4. The effects of several anti-inflammatory drugs on the biosynthesis of leukotriene B_4 were also discussed. The results showed that aspirin, indomethacin, Yacheng Xikang, hydrocortisone and musk first band leukotriene B 4 biosynthesis few effects, and flufenamic acid can inhibit the formation of leukotriene B 4, suggesting that it May be a 5-lipoxygenase inhibitor.
