以切成2mm左右的长筒石蒜幼嫩叶片薄片作为外植体,研究了不同激素组合以及不同接种方式对其愈伤组织诱导的影响,以期获得长筒石蒜叶片愈伤诱导的最佳条件。结果表明:在1/2MS+NAA 0.5mg/L+TDZ 2.0mg/L+蔗糖30g/L+土豆粉3g/L培养基中,愈伤诱导率最高可达90.0%,TDZ诱导愈伤效果明显优于2,4-D;采用叶片垂直插入培养基的接种方式,培养30d后获得愈伤诱导率最高为83.3%。
In order to obtain the optimal conditions for callus induction of leaves of Lycoris radiata, the young leaves of Lycoris radiatus about 2mm were used as explants to study the effects of different combinations of hormones and different inoculation methods on their callus induction. The results showed that the callus induction rate was up to 90.0% in the medium of 1 / 2MS + NAA 0.5mg / L + TDZ 2.0mg / L + sucrose 30g / L + potato starch 3g / L, 2,4-D. The highest callus induction rate reached 83.3% after cultured for 30 days by inoculating the leaves vertically into the medium.
