目的 :采用RP -HPLC法测定贯叶连翘片剂中有效成分金丝桃素 (hypericin)和金丝桃苷 (hyperoside)的含量。方法 :分别使用ODS和ODS - 2色谱柱 ,流动相分别是 1 5 6%磷酸二氢钠 (用磷酸调 pH至 2 1) -甲醇 -醋酸乙酯 ( 1∶4∶1)及甲醇 - 0 5 %磷酸 ( 4 5∶5 5 ,用三乙胺调 pH至 3 0 ) ,检测波长分别为 5 90nm ,3 60nm。结果 :加样回收率分别为 98 3 2 % (n =5 ) ,96 0 4 % (n =5 )。金丝桃素的进样量为 0 0 0 98~ 0 16μg、金丝桃苷的进样量为 0 4 3~ 3 4 2 μg时 ,线性关系良好。结论 :本方法有良好的重现性及精密度
Objective: To determine the contents of hypericin and hyperoside in Hypericum perforatum L. by RP-HPLC. METHODS: ODS and ODS-2 columns were used, the mobile phases were 156% sodium dihydrogen phosphate (pH adjusted to 2 1 with phosphoric acid) - methanol-ethyl acetate (1:4:1) and methanol - 0 5 % phosphoric acid (4 5: 55, adjusted to pH 30 with triethylamine) and the detection wavelengths were 5 90 nm and 3 60 nm, respectively. Results : The recovery rates of the samples were 98 3 2 % (n = 5) and 96 0 4 % (n = 5), respectively. The linear relationship was good when the injection amount of hypericin was 0 0 0 98 - 0 16μg, and the amount of hyperoside was 0 4 3 - 34 2 μg. Conclusion : This method has good reproducibility and precision
