乳猪肝胶原水解物的制备及其对荷瘤小鼠肿瘤生长影响的实验研究

目的 :从乳猪肝中提取可溶性胶原 ,经水解后制备肝胶原水解物 (HCH) ,观察其对荷瘤小鼠S180纤维肉瘤和HepA肝瘤增殖的影响。方法 :从乳猪肝中用生化方法提取可溶性胶原 ,经胃蛋白酶水解制备HCH ;采用SDS -PAGE电泳和激光飞行时间质谱测定其分子量。设立生理盐水阴性对照和阿霉素阳性对照组 ,观察HCH对荷瘤昆明小鼠S180纤维肉瘤和HepA肝瘤增殖的影响。结果 :电泳结果显示 ,HCH的分子量范围为 3 494~ 8 70 2kD ,质谱结果显示 ,其分子量为 2 313~ 8 80 9kD。其对荷瘤小鼠S180纤维肉瘤生长的抑制呈剂量相关性 ,2 5、5 0、10 0mg·kg-1·d-1的抑制率为 5 3 87%、76 5 3 %、5 8 2 0 % ,而阿霉素 (2mg·kg-1·d-1)阳性对照组的抑制率为 6 2 41% ;HCH能剂量依赖性地抑制HepA肝瘤生长 ,2 5、5 0、10 0、2 0 0mg·kg-1·d-1的抑制率为 43 2 6 %、5 5 0 6 %、74 71%、40 47%。结论 :经此方法制备HCH ,其分子量在 2~ 9kDa之间 ,能抑制荷瘤小鼠肿瘤的生长。 OBJECTIVE: To extract soluble collagen from sucking pig liver and hydrolyze it to prepare hepatic collagen hydrolyzate (HCH) and observe its effect on the proliferation of S180 fibrosarcoma and HepA hepatoma in tumor-bearing mice. Methods: The soluble collagen was extracted from the liver of suckling pig by biochemical method, and then hydrolyzed by pepsin to prepare HCH. The molecular weight of HCH was determined by SDS-PAGE and laser-time of flight mass spectrometry. The normal saline control group and doxorubicin positive control group were set up to observe the effect of HCH on the proliferation of S180 fibrosarcoma and HepA hepatoma in tumor-bearing Kunming mice. Results: The results of electrophoresis showed that the molecular weight of HCH ranged from 3 494 to 8 70 2 kD. The mass spectrum showed that the molecular weight of HCH ranged from 2 313 to 8 80 9 kD. Its inhibitory effect on the growth of S180 fibrosarcoma in mice was dose-dependent. The inhibitory rates of 25, 50, 100 mg · kg-1 · d-1 were 53.77%, 76.53%, 582 0%, while the rate of inhibition of adriamycin (2mg · kg-1 · d-1) positive control group was 6241%; HCH dose-dependently inhibited the growth of HepA hepatoma, 25.5% The inhibition rates of 200 mg · kg-1 · d-1 were 43 2 6%, 5 50 6%, 74 71% and 40 47%. Conclusion: HCH prepared by this method has a molecular weight between 2 and 9 kDa, which can inhibit tumor growth in tumor-bearing mice.
