通过前人的总结和自身实验,创建和改良兰州百合的鳞片组培快繁体系,并初步进行以Oryzalin为诱导试剂的多倍体诱导实验。结果表明:鳞片消毒以75%酒精浸泡30 s、2%次氯酸钠浸泡20 min与0.1%升汞浸泡20 min,效果最佳;不定芽增殖以MS+6.25 g/L琼脂+80.00 g/L蔗糖+1.50 mg/L 6-BA+0.10 mg/L NAA作为培养基最佳;组培苗生根以MS+6.25 g/L琼脂+80.00 g/L蔗糖+0.20 mg/L NAA作为培养基最佳;在0.002%Oryzalin溶液浸泡24 h的处理中,发现疑似多倍体,有待进一步确定。
Through the summary of predecessors and their own experiments, we established and improved the rapid tissue culture system of Lanzhou lily by polyploid induction with Oryzalin as the induction reagent. The results showed that the best effect was obtained when soaked in 75% alcohol for 30 s, soaked in 2% sodium hypochlorite for 20 min and 0.1% mercuric chloride for 20 min. The proliferation of adventitious buds was best when MS + 6.25 g / L agar +80.00 g / L sucrose + 1.50 mg / L 6-BA + 0.10 mg / L NAA was the best culture medium. The optimal culture medium was MS + 6.25 g / L agar +80.00 g / L sucrose +0.20 mg / L NAA. In the treatment of 0.002% Oryzalin solution soaked for 24 h, the suspected polyploid was found to be further determined.
