目的观察芪珀生脉颗粒对房颤大鼠模型cAMP、cGMP、cAMP/cGMP的影响。方法采用乙酰胆碱-氯化钙混合液通过尾静脉给药建立房颤大鼠模型。根据预实验房颤造模死亡情况估计,从90只SD大鼠中随机抽取8只作为正常组,其余大鼠分为模型组、稳心颗粒组、维拉帕米组、芪珀生脉颗粒大剂量组(大剂量组)、芪珀生脉颗粒中剂量组(中剂量组)、芪珀生脉颗粒小剂量组(小剂量组)。治疗4周后,测定各组大鼠血浆cAMP、cGMP,计算cAMP/cGMP并进行统计学分析。结果各组间cAMP水平差异无统计学意义(P>0.05);模型组cGMP水平低于除小剂量外的各组(P<0.05),其中与正常组、维拉帕米组、大剂量组间差异有统计学意义(P<0.01);各组cGMP水平均高于小剂量组(P<0.05),其中正常组、维拉帕米组、大剂量组间差异有统计学意义(P<0.01);模型组cAMP/cGMP水平高于正常组、维拉帕米组、大剂量组,组间差异有统计学意义(P<0.05)。结论芪珀生脉颗粒能够有效升高房颤大鼠cGMP,降低cAMP/cGMP。
Objective To observe the effects of Qiaolian Shengmai Granule on cAMP, cGMP and cAMP / cGMP in atrial fibrillation rat model. Methods Atrial fibrillation rat model was established by intravenous infusion of acetylcholine-calcium chloride. According to the pre-experimental atrial fibrillation model of death estimates, 90 SD rats were randomly selected as the normal group of eight, the remaining rats were divided into model group, Wenxin Granule group, verapamil group, Qi Pei Sheng Mai particles High-dose group (high-dose group), Qibei Shengmai granule medium dose group (middle dose group) and Qibei Shengmai Granules small dose group (low dose group). After 4 weeks of treatment, the plasma levels of cAMP and cGMP in each group were measured and cAMP / cGMP was calculated and statistically analyzed. Results There was no significant difference in cAMP levels between groups (P> 0.05). The level of cGMP in model group was lower than that in other groups (P <0.05) except normal group, verapamil group, high dose group (P <0.01). The levels of cGMP in each group were significantly higher than those in the low-dose group (P <0.05), and there was a significant difference between the normal group, verapamil group and the high-dose group (P < 0.01). The level of cAMP / cGMP in the model group was higher than that in the normal group. The verapamil group and the high dose group had statistically significant differences (P <0.05). Conclusion Qibe Shengmai Granules can effectively increase cGMP and decrease cAMP / cGMP in atrial fibrillation rats.
