重组人白细胞介素18的纯化及其生物学活性研究

目的 :纯化rhIL 18并对其生物学功能进行初步研究。方法 :采用优化的包涵体提取技术和SephacrylS 2 0 0分子筛层析纯化rhIL 18,利用MTT染色法研究rhIL 18对NK细胞促杀伤作用和对外周血单个核细胞 (PBMC)的促增殖作用 ,采用半定量RT PCR方法研究rhIL 18诱导IFN γ基因的表达情况。结果 :得到纯度达 96 %的纯品rhIL 18,复性的rhIL 18可以明显地促进PBMC增殖 ,增强NK细胞细胞毒活性。结论 :得到纯度较高、具有较好生物学功能的rhIL 18,可用于进一步的研究 Objective: To purify rhIL 18 and study its biological function. Methods: The optimized inclusion body extraction technology and purification by Sephacryls 2000 molecular sieve chromatography rhIL 18, the use of MTT staining to study the role of rhIL 18 promote NK cell cytotoxicity and peripheral blood mononuclear cells (PBMC) to promote proliferation, using Semiquantitative RT-PCR method was used to study the expression of IFN-γ induced by rhIL-18. Results: Pure rhIL 18 with purity of 96% was obtained. Refolded rhIL 18 could obviously promote the proliferation of PBMC and enhance the cytotoxic activity of NK cells. Conclusion: The rhIL 18 with higher purity and better biological function can be used for further research
