目的建立苏氏接骨胶囊中当归和川芎的定性鉴别和总阿魏酸的定量检测方法。方法采用薄层色谱法对苏氏接骨胶囊中的当归和川芎进行定性鉴别,采用高效液相色谱法对阿魏酸进行定量测定。色谱柱为Thermo C18(250mm×4.6mm,5μm)柱,流动相为乙腈-1%醋酸溶液(20:80),体积流量1.0mL/min,柱温25℃,检测波长320nm。结果薄层色谱清晰,分离良好,阴性无干扰。阿魏酸在0.42~2.24μg线性关系良好(r=0.9999),平均回收率为99.26%,RSD为1.27%。结论所建立的方法准确、快速、简便、重现性好,可作为苏氏接骨胶囊的质量控制方法。
Objective To establish the qualitative identification of Angelica sinensis and Chuanxiong in Suji Jiegu capsule and the quantitative determination of total ferulic acid. Methods TLC and HPLC were used to identify Angelica sinensis and Chuanxiong rhizome. The contents of ferulic acid were determined by high performance liquid chromatography (HPLC). The column was Thermo C18 (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-1% acetic acid solution (20:80), the volume flow rate was 1.0 mL / min, the column temperature was 25 ℃ and the detection wavelength was 320 nm. Results TLC was clear, good separation, negative non-interference. Ferulic acid had a good linearity (r = 0.9999) between 0.42 and 2.24 μg with an average recovery of 99.26% and a RSD of 1.27%. Conclusion The established method is accurate, rapid, simple and reproducible. It can be used as quality control method of Suji Jiegu capsule.
