目的:了解DC在胃癌胃黏膜中的功能状态,探讨DC用于胃癌等实体瘤防治的可能性,旨在为胃癌的治疗探索一条新的途径. 方法:利用致敏DC治疗胃癌,以观察其对荷胃癌小鼠的免疫治疗效果. 结果:荷胃癌小鼠经胃癌细胞RNA致敏的DC治疗后,小鼠腹水产生率、肿瘤转移率、动物的存活率、平均瘤体质量等指标分别为75%(6/8)、25%(2/8)、75%(6/8)、(2.04±0.33 g),均优于对照组(P<0.01);局部组织中IL-12、IL-18 和IFN γ的基因表达水平都明显升高(P<0.01);肿瘤局部CD4+、CD8+细胞数量分别为0.71±0.25/μm2和0.67±0.22/μm2, P<0.05),TIL细胞毒活性明显增强. 结论:胃癌RNA致敏的DC,能促进胃癌抗原的提呈并启动和增强局部的抗瘤免疫功能,改善临床症状和生存质量, 对荷胃癌小鼠具有一定的免疫治疗效果.
OBJECTIVE: To understand the functional status of DC in gastric mucosa of gastric cancer and explore the possibility of its application in the prevention and treatment of solid tumors such as gastric cancer, and to explore a new approach for the treatment of gastric cancer.Methods: On mice bearing gastric cancer.Results: After gastric cancer cells were sensitized with DCs, the incidence of ascites, the rate of tumor metastasis, the survival rate of animals and the mean tumor mass were all 75% (6/8), 25% (2/8), 75% (6/8) and (2.04 ± 0.33 g) respectively, which were all better than the control group (P <0.01) (P <0.01). The numbers of CD4 + and CD8 + cells in local tumor were 0.71 ± 0.25 / μm2 and 0.67 ± 0.22 / μm2 respectively (P <0.05), and the TIL cytotoxicity was significantly higher Enhanced.Conclusion: RNA-sensitized DCs of gastric cancer can promote the presentation of gastric cancer antigens and start up and enhance the local anti-tumor immune function, improve the clinical symptoms and quality of life, and have certain immunotherapy effect on gastric cancer-bearing mice.
