应用角膜中期保存法(K液、CSM、Dexsol及Optisol)对85只动物角膜及5只人角膜进行保存试验,保存时间为3、8、10、14及20天。应用0.25%锥蓝及0.2%茜素红双染色,保存10天以内的动物及人角膜内皮细胞完整性良好,14天的供体角膜部分出现水肿,内皮细胞丢失率为5%~10%,保存20天的供俸角膜大部分出现水肿,内皮细胞丢失率在30%~40%。对保存8及10天之动物角膜进行电镜检查,其超微结构基本正常。应用中期保存法保存1~10天的476只人角膜供体行临床穿透性角膜移植术,术后除6例发生供体失败外,其他病例术后植片均透明,术后随访3个月~3年,植片透明率为81%,内皮细胞密度为810~2640个/mm~2。
85 corneas and 5 human corneas were preserved in the cornea mid-term preservation (K solution, CSM, Dexsol and Optisol) for 3, 8, 10, 14 and 20 days. Double staining with 0.25% conical blue and 0.2% alizarin red showed that the integrity of corneal endothelial cells in animals and human corneas preserved within 10 days was good. Edema was observed in donor cornea for 14 days. The endothelial cell loss rate was 5% -10% Save 20 days for most of corneal necrosis edema, endothelial cell loss rate of 30% to 40%. The cornea preserved for 8 and 10 days underwent electron microscopy and its ultrastructure was basically normal. 476 corneal donors who underwent 1 ~ 10 days postoperative preservation were treated with clinical penetrating keratoplasty. Six cases had donor failure after operation, and the other cases were transplanted transparently after operation. Three cases were followed up Month ~ 3 years, the transparency of the graft was 81%, endothelial cell density was 810 ~ 2640 / mm ~ 2.
