为了观察复方浙贝颗粒(CZBG)联合阿霉素对K562/A02移植瘤细胞膜耐药相关蛋白P-gp、MRP、LRP表达的影响,将K562/A02细胞接种于BALB/c-nu裸小鼠腋前皮下以构建MDR移植瘤动物模型,给予复方浙贝颗粒灌胃联合阿霉素腹腔注射,实验结束后处死小鼠,剥离肿瘤,将瘤块组织制成切片,通过免疫组织化学结合Image ProP lus6.0免疫组织化学彩色图象分析,观察复方浙贝颗粒联合阿霉素对K562/A02移植瘤组织细胞膜P-gp、MRP、LRP表达的影响。结果表明:与阿霉素组相比,CZBG各剂量组移植瘤细胞膜所表达P-gp、MRP的积分光密度值(IOD)明显降低(p<0.05),而未见明显LRP表达。结论:CZBG能协同阿霉素抑制K562/A02移植瘤组织细胞膜P-gp、MRP表达。
To observe the effects of CZBG and doxorubicin on the expression of P-gp, MRP and LRP in K562 / A02 tumor cells, K562 / A02 cells were inoculated into BALB / c nu mice The axillary anterior subcutaneous tissue was used to construct the MDR xenograft model. The mice were injected intraperitoneally with Compound ZheBai Granules and doxorubicin. After the experiment, the mice were sacrificed and the tumor was stripped. The tumor mass was made into sections. Immunohistochemistry and Image ProP lus6.0 immunohistochemistry color image analysis to observe the effect of compound ZheBi granule combined with doxorubicin on the expression of P-gp, MRP and LRP in K562 / A02 transplanted tumor cell membrane. The results showed that the integral optical density (IOD) of P-gp and MRP in CZBG transplanted tumor cells was significantly lower than that in doxorubicin group (p <0.05), but no obvious LRP expression. Conclusion: CZBG can cooperate with doxorubicin to inhibit the expression of P-gp and MRP in the cell membrane of K562 / A02 xenografts.
