目的:通过化学成分和药效学指标的研究,确定赤芍、大黄药对的最佳配伍比例。方法:用HPLC对赤芍、大黄药对提取物中成分(芍药苷、芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚)与药效结合进行分析。采用色谱柱为Phenomsil BDS C18柱(250 mm×4.6 mm,5μm);检测大黄指标性成分流动相为甲醇-0.1%磷酸(85∶15),检测波长254nm;条件2:检测赤芍指标性成分流动相为乙腈-水(14∶86),检测波长为230 nm;流速均为1.0 m L·min-1,柱温25℃。药效测定采用毛细管法测定凝血时间。结果:样品出膏率、HPLC指标性成分、药效评分均以3∶1最高。结论:确定了赤芍、大黄药对的最佳配伍比例为3∶1。
OBJECTIVE: To determine the optimal compatibility of Radix Paeoniae Rubra and Rhubarb by the study of chemical composition and pharmacodynamic parameters. Methods: The constituents of paeoniflorin and rhubarb were extracted by HPLC (paeoniflorin, aloe emodin, rhein, emodin, chrysophanol and physcion) and their pharmacodynamics were analyzed. The chromatographic column was Phenomsil BDS C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase of rhubarb was methanol-0.1% phosphoric acid (85:15) and the detection wavelength was 254 nm. Condition 2: The mobile phase consisted of acetonitrile-water (14:86) and the detection wavelength was 230 nm. The flow rate was 1.0 m L · min-1 and the column temperature was 25 ℃. Pharmacodynamics Determination of clotting time by capillary method. Results: The rate of sample ointment, HPLC index components, efficacy scores were the highest 3: 1. Conclusion: The optimum proportion of Radix Paeoniae Rubra and Rhubarb is 3:1.
