目的 :电镜下观察不同时间、不同超剂量组 RU486 对大鼠卵巢中细胞亚显微结构的影响 ,从形态学角度研究 RU486 避孕机制。方法 :给实验大鼠灌服不同超剂量 RU486 ,于不同时间取卵巢组织 ,制作电镜标本并观察。结果 :实验各组生长卵泡发育不良或闭锁 ,未见成熟卵泡和黄体 ,闭锁卵泡、间质腺增多 ,卵泡细胞、间质腺细胞内含大量脂滴、空泡、滑面内质网 ( SER)及次级溶酶体 ( s Ly) ,巨噬细胞、变异颗粒细胞内出现残余体和聚集成团的溶酶体 ( L y) ,其超微结构改变随用药时间延长而加重。结论 :使用不同超剂量 RU486 可抑制卵泡发育成熟及黄体形成 ,并对卵巢内部分细胞的细胞器有一定的损害。
OBJECTIVE: To observe the effect of RU486 on ovarian micro-structure in rat ovaries at different time and different doses, and to study the mechanism of RU486 contraception from the morphological point of view. Methods: Different doses of RU486 were given to the experimental rats, ovarian tissues were taken at different times, and the electron microscopy specimens were made and observed. Results: Follicles and interstitial glands in follicular cells and interstitial glands contained a large number of lipid droplets, vacuoles and smooth endoplasmic reticulum (SER) in all experimental groups. And secondary lysosomes (s Ly), macrophages, mutant granulosa cells and the accumulation of lysosomes (Ly), the ultrastructural changes with the use of drugs for an extended period of aggravate. Conclusion: RU486 can inhibit the development of ovarian follicles and the formation of corpus luteum, and some damage to organelles of some cells in the ovary.
