目的 扩增与人成骨肉瘤细胞特异结合的鼠源性单克隆抗体轻链基因 ,并对其基因序列进行分析 .方法 用RT- PCR方法从分泌鼠源抗人成骨肉瘤单克隆抗体的杂交瘤细胞 ( 7F6 B)扩增出抗体轻链基因 ,克隆到 p GEM- T载体 ,以全自动测序仪测定其轻链基因序列并对其结构进行分析 .结果 7F6 B轻链基因由 6 5 6 bp组成 ,编码 2 18个氨基酸 ,序列分析证实该基因可变区序列符合小鼠 Ig轻链基因的氨基酸序列特征 .结论 获得的鼠源性抗成骨肉瘤抗体轻链基因为κ 家族 ,该结果为进一步探讨人成骨肉瘤的免疫治疗研究奠定了基础
Objective To amplify the murine monoclonal antibody light chain gene that specifically binds to human osteosarcoma cells and analyze its gene sequence.METHODS The hybridization of murine anti-human osteosarcoma monoclonal antibody The light chain gene was amplified from the tumor cells (7F6 B) and cloned into p GEM-T vector, and its light chain gene sequence was determined by automated sequencer and its structure was analyzed.Results The 7F6 B light chain gene was composed of 6 5 6 bp, encoding 2 18 amino acids.The sequence analysis confirmed that the variable region of this gene conformed to the amino acid sequence of mouse Ig light chain gene.Conclusion The murine anti-osteosarcoma antibody light chain gene is a kappa family, This study lays the foundation for further research on the immunotherapy of human osteosarcoma
