目的:对仙茅多糖进行分离纯化,并对其结构进行分析,为仙茅多糖的进一步应用提供理论基础。方法:仙茅根茎经水提、醇沉、冷冻干燥得仙茅粗多糖(XMP),经DEAE-纤维素柱和Sepharose CL-6B凝胶柱色谱纯化得纯多糖XMP-1,采用凝胶渗透色谱、紫外光谱、红外光谱、气质联用等技术对XMP-1的结构进行分析。结果:仙茅多糖由甘露糖、葡萄糖、半乳糖3种单糖组成,其摩尔比为81.2∶5.7∶1,多糖质量分数为91.8%,相对分子质量为3 031 Da,不含核酸、蛋白质和色素。具有吡喃特征吸收峰,在水溶液分子中的分子构象可能为自然卷曲结构。结论:仙茅纯多糖是一种分子量较小的中性杂多糖。
OBJECTIVE: To isolate and purify Curculigo lactiflora polysaccharides and analyze its structure, providing the theoretical basis for the further application of curcumin. Methods: Rhizoma curculigo was subjected to water extraction, alcohol precipitation and freeze-drying to obtain crude polysaccharides XMP. The pure polysaccharide XMP-1 was purified by DEAE-cellulose column and Sepharose CL-6B gel column chromatography. Chromatography, UV spectroscopy, infrared spectroscopy, GC-MS and other technologies to analyze the structure of XMP-1. Results: Curculin was composed of three kinds of monosaccharides of mannose, glucose and galactose, the molar ratio was 81.2: 5.7: 1, the polysaccharide content was 91.8% and the relative molecular mass was 3 031 Da. The content of nucleic acid, protein and pigment. Has a characteristic absorption peak of pyran and the molecular conformation in the molecule of aqueous solution may be a natural curly structure. Conclusion: Curculigo pure polysaccharide is a low molecular weight neutral heteropolysaccharide.
