目的探讨正常妊娠和妊娠期甲状腺功能减退症胎盘中核转录因子κB(NF-κB)以及环氧合酶-2(COX-2)表达的意义。方法采用免疫组织化学二步法检测正常胎盘(35例)和妊娠期甲状腺功能减退症胎盘(60例)组织中NF-κB和COX-2的表达水平。结果在正常妊娠胎盘组织的绒毛滋养细胞、绒毛间质细胞和血管内皮细胞胞浆中均可见NF-κB和COX-2的阳性表达,多为弱阳性和中等强度阳性表达;在妊娠期甲状腺功能减退症胎盘组织中NF-κB和COX-2的表达部位与正常组相同,但着色更深,多为强阳性表达;妊娠期甲状腺功能减退症胎盘组织NF-κB和COX-2表达呈正相关(r=0.650,P<0.05)。结论妊娠期甲状腺功能减退症胎盘组织中NF-κB和COX-2表达增多可能是妊娠期甲状腺功能减退症病理生理过程中重要的内在因子。
Objective To investigate the significance of the expression of nuclear factor-kappa B (NF-κB) and cyclooxygenase-2 (COX-2) in the placenta of normal pregnancy and hypothyroidism during pregnancy. Methods The expressions of NF-κB and COX-2 in normal placenta (35 cases) and hypothyroidism in pregnancy (60 cases) were detected by immunohistochemical two-step method. Results The positive expression of NF-κB and COX-2 were found in the placenta of normal pregnancy with villus trophoblast, villus mesenchymal cells and vascular endothelial cells, most of which were weakly positive and moderately positive. In thyroid function during pregnancy The expression of NF-κB and COX-2 in placenta of hypothyroidism was the same as that in normal group, but the expression was deeper and more strongly positive. The expression of NF-κB and COX-2 in placenta of hypothyroidism during pregnancy was positively correlated = 0.650, P <0.05). Conclusion The increased expression of NF-κB and COX-2 in placenta during hypothyroidism during pregnancy may be an important intrinsic factor in the pathophysiology of hypothyroidism during pregnancy.
