目的:探讨bFGF基因转染骨髓基质干细胞的方法及可行性。方法:构建bFGF基因真核表达质粒,用脂质体法介导转染骨髓基质细胞,通过免疫组织化学、RT-PCR及Western blot方法检测bFGF基因转染骨髓基质细胞的成功性。结果:bFGF基因成功转染大鼠骨髓基质细胞,并能持续稳定地分泌bFGF蛋白。结论:bFGF基因可以转染骨髓基质细胞,并能在骨髓基质细胞内稳定表达。
Objective: To investigate the feasibility and feasibility of bFGF gene transfection into bone marrow stromal stem cells. Methods: The eukaryotic expression plasmid of bFGF gene was constructed and transfected into bone marrow stromal cells by liposome. The success of bFGF transfection into bone marrow stromal cells was detected by immunohistochemistry, RT-PCR and Western blot. Results: The bFGF gene was successfully transfected into rat bone marrow stromal cells and bFGF protein was secreted continuously and stably. Conclusion: The bFGF gene can be transfected into bone marrow stromal cells and stably expressed in bone marrow stromal cells.
