目的:探讨油酸型急性呼吸窘迫综合征(ARDS)大鼠心肌组织中白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)含量及其mRNA的表达在心肌损伤过程中的作用。方法:Wistar大鼠32只,随机分为4组,采用油酸复制ARDS大鼠模型,观察对照组、油酸(OA)注射后2h(OA2h组)、4h(OA4h组)、6h(OA6h组)组肺的湿/干重、心肌组织的病理改变及血清心肌酶谱(AST、CK、CK-MB、LDH)。用放射免疫分析法检测血清及心肌组织中IL-1β、IL-10的含量,以RT-PCR法检测心肌组织中IL-1β、IL-10mRNA的表达情况。结果:OA各组肺湿/干重均高于C组(P<0.05)。OA各组AST以及OA2h、OA4h组CK、CK-MB、LDH均高于对照组(P<0.05)。OA6h组血清及心肌组织中的IL-1β含量显著升高(P<0.01);OA2h组血清IL-10含量明显高于其它各组(P<0.01),OA6h组心肌组织中IL-10含量明显高于对照组及OA2h组(P<0.01)。OA各组心肌组织IL-1βmRNA表达均高于对照组(P<0.01);OA6h组IL-10mRNA表达有所升高(P<0.01)。结论:油酸型ARDS大鼠并发心肌损伤可能与心肌组织中IL-1β、IL-10含量的增高、mRNA表达的增强有关。
Objective: To investigate the expression of interleukin-1β (IL-1β), interleukin-10 (IL-10) and its mRNA in myocardium of acute exogenic respiratory distress syndrome (ARDS) In the role. Methods: Thirty-two Wistar rats were randomly divided into 4 groups. The rats in the control group were treated with oleic acid for 2 days (OA2h group), 4h (OA4h group), 6h (OA6h group) ) Group lung wet / dry weight, myocardial tissue pathological changes and serum myocardial enzymes (AST, CK, CK-MB, LDH). The levels of IL-1β and IL-10 in serum and myocardium were detected by radioimmunoassay, and the expression of IL-1β and IL-10 mRNA in myocardium was detected by RT-PCR. Results: The wet / dry weights of lung in OA group were higher than those in C group (P <0.05). The levels of CK, CK-MB and LDH in OA and OA2h and OA4h groups were higher than those in control group (P <0.05). The levels of IL-1β in serum and myocardial tissue of OA6h group were significantly increased (P <0.01), while the levels of IL-10 in OA2h group were significantly higher than those in other groups (P <0.01) Higher than the control group and OA2h group (P <0.01). The expression of IL-1βmRNA in myocardium of OA group was higher than that of the control group (P <0.01). The expression of IL-10mRNA in OA6h group was increased (P <0.01). CONCLUSION: Myocardial injury induced by oleic acid in ARDS rats may be related to the increase of IL-1β and IL-10 content and the increase of mRNA expression in myocardium.
