目的构建20种常见高低危型HPV编码蛋白进化树,分析其进化关系。方法从NCBI数据库中获取常见20种高低危型HPV的8种编码蛋白完整氨基酸序列,通过MEGA6.0软件中邻位连接法(neighbor-joining,NJ)构建系统亲缘进化树,进行进化分析。结果低危型HPV6、11和42亚型单独处于一分支上,并与高危型HPV分开。在高危型中,HPV18、39、45、59和68亚型,HPV16、31和35亚型以及HPV33、52和58亚型的HPV基因组8种编码蛋白均分别聚集在同一支上,而高危型HPV73的8种编码蛋白在进化树上的位置均不固定。在构建的E2和E6蛋白进化树中分歧支较多,结果较不可信。结论在蛋白进化树中常见低危型HPV均单独处于一支,且与高危型HPV分开,12个高危型HPV分布于3个不同分支或位置不固定。
Objective To construct 20 kinds of common high and low risk HPV coding protein phylogenetic tree and analyze its evolutionary relationship. Methods The complete amino acid sequences of eight kinds of encoded proteins of 20 kinds of high and low risk HPVs were obtained from NCBI database. Phylogenetic tree of phylogenetic tree was constructed by neighbor-joining (MEGA) 6.0 software and phylogenetic tree was used for phylogenetic analysis. Results The low-risk HPV6, 11 and 42 subtypes were separated on a single branch and separated from high-risk HPV. In the high-risk type, the HPV18, 39, 45, 59 and 68 subtypes, the HPV16, 31 and 35 subtypes, and the HPV genotypes 15, 52 and 58 were all clustered together on the same branch. The eight kinds of HPV73 encoded proteins are not fixed on the phylogenetic tree. In the constructed E2 and E6 protein phylogenetic tree divergence more, the result is less credible. Conclusion In the phylogenetic tree, the common low-risk HPVs are all located in one and separate from the high-risk HPVs. Twelve high-risk HPVs are located in three different branches or are not fixed.
