目的:观察体外诱导的自体白血病树突状细胞(dendriticcell,DC)疫苗体内回输的安全性,及对体内免疫状态的影响。方法:20例白血病患者接受体外诱导的自体白血病树突状细胞疫苗的静脉回输。留取患者血清及外周血细胞分别行流式小球微阵列术(cytokinebeadarray,CBA)分析血清Th1/Th2因子IFNγ、IL2、IL4、IL6、IL10和TNFα浓度,流式细胞学(flowcytometricmethod,FCM)分析外周血T细胞亚群表达变化。结果:体外诱导的自体白血病DC疫苗回输后,临床未观察到明显的不良反应。第一次DC疫苗回输前及回输后24h比较,血清Th1/Th2因子浓度均升高,其中IL2、IL4、IL6和IFNγ升高显著,t值分别为2.576、2.033、2.777和2.053,P值分别为0.01、0.042、0.005和0.040。T亚群分析显示,CD4/CD8比值及NK比率升高,t值为2.040和1.988,P值为0.041和0.043。结论:自体白血病DC疫苗静脉回输安全可行,并可刺激患者体内的免疫反应,增强患者的免疫功能。
OBJECTIVE: To observe the safety of dendriticcell (DC) vaccine in vitro and its effect on immune status in vitro. Methods: Twenty leukemia patients received intravenous infusion of an autologous leukemia dendritic cell vaccine induced in vitro. Serum and peripheral blood cells were collected and analyzed for serum Th1 / Th2 cytokines IFNγ, IL2, IL4, IL6, IL10 and TNFα by flow cytometry (FCM) and flow cytometry (FCM) Changes of T cell subsets expression in peripheral blood. RESULTS: No adverse reactions were observed clinically after the DC vaccination of autologous leukemia was induced in vitro. The levels of serum Th1 / Th2 factors increased before and after the first vaccination of DC vaccine, and the levels of IL2, IL4, IL6 and IFNγ were significantly increased, with t values of 2.576, 2.033, 2.777 and 2.053, respectively The values were 0.01, 0.042, 0.005 and 0.040, respectively. T-subgroup analysis showed that CD4 / CD8 ratio and NK ratio increased, t values were 2.040 and 1.988, P values were 0.041 and 0.043. Conclusion: Autologous leukemia DC vaccine is safe and feasible for intravenous infusion and stimulates the immune response in patients and enhances the immune function of patients.
