为诱导胚胎干细胞向软骨细胞分化,采用胚胎干细胞与成熟软骨细胞共培养的方法以提供软骨诱导微环境.小鼠胚胎干细胞经初步分化,形成类胚体后,从中分选出Flk-1阳性细胞,其与猪关节软骨细胞混合后,接种于可降解材料支架,并植入裸鼠皮下.4周后经组织学检查及Ⅱ型胶原抗体免疫荧光检查证实,细胞材料复合物形成软骨组织.经小鼠主要组织相容性抗原染色,证实其中部分软骨细胞源自于小鼠胚胎干细胞.本研究建立了胚胎干细胞定向诱导分化的新方法.
To induce differentiation of embryonic stem cells into chondrocytes, co-culture of embryonic stem cells with mature chondrocytes was used to provide cartilage-induced microenvironment.After embryonic stem cells were initially differentiated to form embryoid bodies, Flk-1 positive cells , Which was mixed with porcine articular chondrocytes and inoculated on the scaffolds of biodegradable material and subcutaneously implanted into nude mice.After 4 weeks, histological examination and type II collagen antibody immunofluorescence examination confirmed that the cellular material composites formed cartilage tissue. The major histocompatibility antigens of mice were stained to confirm that some of the chondrocytes originated from the mouse embryonic stem cells.This study established a new method of directed differentiation of embryonic stem cells.
