目的采用高速逆流色谱法,制备延胡索中的4个主要生物碱。方法应用高速逆流色谱仪,以氯仿-甲醇-0.1mol/L盐酸(4∶3∶2,V/V/V)或(4∶3∶4,V/V/V)的上相为固定相,下相为流动相,组成溶剂系统对延胡索的生物碱进行分离纯化。结果分离得到4个生物碱,分别为非洲防己碱、黄连碱、巴马汀、去氢紫堇碱,纯度依次为91%、94%、92%、97%。结论建立的高速逆流色谱法,可用于延胡索中主要生物碱的制备,该方法快速、简便,产物得率和纯度较高。
Objective To prepare four major alkaloids from Corydalis yanhusuo by high-speed countercurrent chromatography. Methods The high-speed countercurrent chromatograph was used with the upper phase of chloroform-methanol-0.1mol / L hydrochloric acid (4: 3: 2, V / V / V) or (4: 3: 4, V / V / V) , The next phase as the mobile phase, the solvent system composed of corydalis alkaloids were isolated and purified. Results Four alkaloids were isolated, which were respectively tetrandrine, coptisine, palmatine and corydaline with the purity of 91%, 94%, 92% and 97%, respectively. Conclusion The established high-speed countercurrent chromatography can be used for the preparation of major alkaloids in Corydalis yanhusuo. The method is rapid and simple, and the yield and purity of the product are high.
